چکیده

Background: The immunopathogenesis of pediatric multiple sclerosis (MS) is not well understood. Methods: We studied the cytokine profile in pre-treatment serum specimens of 19 pediatric MS patients, 25 adult MS patients, and 22 ageand gender-matched pediatric healthy controls. In addition to IL-2, IL-12p40, IL-12p70, IL-18, IL-23, IL-6, TNF-α, TGF-β-1, IFN-γ, IL-17A, IL-21, IL-10, IL-4, IL-5, IL-13, and GM-CSF, we measured osteopontin and soluble VCAM-I. Results: In children with MS, significantly lower levels of IL-6 were present compared to ageand gender-matched healthy control children (p < 0.05). Moreover, significantly higher levels of osteopontin (p < 0.02) and sVCAM-1 (p < 0.02) and lower levels of IL-6 (p < 0.01) were present, with trends toward lower levels of IL-12p70 (p = 0.074) and IL-17a (p = 0.05) compared to adults with MS. Conclusions: These findings indicate important differences in cytokine signatures in children with MS and suggest an unexpected possible lower inflammatory cytokine profile in children with MS.

Altered Serum Cytokine Profiles in Relapse Phase of Relapsing-Remitting Multiple Sclerosis

Background: Multiple sclerosis (MS) is a demyelinating disease of the central nervous system and cytokines may play a role in the development of MS lesions. Objective: To determine levels of different cytokines in patients with relapsing-remitting MS (RR-MS) compared to healthy controls. Methods: Profiles of pro-inflammatory, Th1-, Th2-, and Th17-related cytokines were compared by quantitative ...

Associations between the gut microbiota and host immune markers in pediatric multiple sclerosis and controls

BACKGROUND As little is known of association(s) between gut microbiota profiles and host immunological markers, we explored these in children with and without multiple sclerosis (MS). METHODS Children ≤18 years provided stool and blood. MS cases were within 2-years of onset. Fecal 16S rRNA gene profiles were generated on an Illumina Miseq platform. Peripheral blood mononuclear cells were isol...

Th1, Th2 and Th17 Cytokine Profile in Patients with Multiple Sclerosis Following Treatment with Rapamycin

Background: Management of multiple sclerosis (MS) is based on the usage of immunosuppressive and immune-modulating medications. Cytokines play an important role in the pathogenesis of MS. Objective: To evaluate the effects of rapamycin on the concentrations of Th1/Th2/Th17 serum cytokines in patients with MS. Methods: Six patients with relapsing remitting MS as a case group and 6 healthy indivi...

P89: The Relationship Between the TNFα of the Microglial Cells and the Multiple Sclerosis

TNFα is an inflammatory cytokine and the caspase and apoptotic processes can be activated by TNFα There are two classes of TNFα (solTNF and tmTNFα) which are important from the receptor aspect, so that solTNFα and tmTNFα can attach to the TNFR1, but tmTNFα just attaches to the TNFR2. Microglia cells are resident immune cells of the central nervous ...

Cytokine profiles show heterogeneity of interferon-β response in multiple sclerosis patients

OBJECTIVE To evaluate serum cytokine profiles for their utility to determine the heterogeneous responses to interferon (IFN)-β treatment in patients with multiple sclerosis (MS). METHODS Patients with relapsing-remitting MS (RRMS) or clinically isolated syndrome receiving de novo IFN-β treatment were included in this prospective, observational study. Number of relapses and changes in disabili...
